Ashour Al-Taqui scored Wadi Degla's first ever goal in the top flight after three minutes to stun one-time Egyptian champions Contractors.
Contractors leveled the score through a goal from Nigerian striker Musa Kabiru in the 39th minute but Degla ran riot to enjoy a night to remember on their league debut.
Syrian striker Abdul-Fattah Agha, who joined Degla in the close season, restored his side's advantage two minutes before the break.
Ahmed Mostafa added the third eight minutes past the hour mark and Ghanaian striker Wisdom Agblexo completed the rout in stoppage time.
Wadi Degla, which are owned by Egyptian businessman Maged Sami, are one of the few privately-owned clubs in the top flight.
Sami also owns Belgian side SK Lierse, who sealed promotion to the first division this season.
Fellow debutants Smouha, who are also playing in the Premier League for the first time in their history, were unlucky though, losing 1-0 to last season's dark horses Military Production.
An own goal from Smouha's newly-signed defender Mohsen Hendawi in the 75th minute gave Production an opening victory.
